Abstract
ABSTRACT Background : Giemsa staining is an essential technique in peripheral blood smear (PBS) examination to analyze blood cell morphology. Conventional methods often lead to reagent waste and uneven results due to stain sedimentation. Chamber stain is an innovative tool designed to improve the effectiveness and efficiency of the staining process. Objectives : To determine the effectiveness and efficiency of repeated use of 15% Giemsa in a Chamber stain for PBS staining compared to the conventional method, and to assess whether repeated use of Giemsa in a Chamber stain affects staining quality. Methods : This study used a pre-experimental design with a posttest-only control group design. Samples were EDTA venous blood from 6th-semester students of the D3 Medical Laboratory Technology Program at Poltekkes Kemenkes Yogyakarta. Two staining methods were applied: conventional and Chamber stain. Microscopic evaluations were conducted by two Medical Laboratory Technologists. Results : The results showed that Chamber stain usage maintained good staining quality up to the 4th day, with a decline observed on the 5th day. This method saved up to 88,33% of Giemsa reagent usage and produced uniform staining without sedimentation. Conclusion : Repeated use of 15% Giemsa with a Chamber stain is effective and efficient. The Chamber stain is proven to be more efficient in terms of reagent usage and staining time compared to the conventional method and can maintain comparable microscopic quality within a certain usage limit. ABSTRAK Latar Belakang : Pewarnaan Giemsa merupakan teknik penting dalam pemeriksaan sediaan apus darah tepi (SADT) untuk menganalisis morfologi sel darah. Metode konvensional sering kali menyebabkan pemborosan reagen dan hasil yang tidak merata karena pengendapan pewarna. Chamber stain merupakan alat inovatif yang dirancang untuk meningkatkan efektivitas dan efisiensi proses pewarnaan. Tujuan : Mengetahui efektivitas dan efisiensi penggunaan Giemsa 15% secara berulang dalam Chamber stain pada pewarnaan SADT dibandingkan metode konvensional, serta mengetahui apakah pemakaian Giemsa secara berulang dalam Chamber stain mempengaruhi kualitas hasil pewarnaan. Metode : Penelitian ini menggunakan desain pra-eksperimental dengan posttest-only control group design. Sampel berupa darah vena EDTA dari mahasiswa semester 6 Prodi D3 Teknologi Laboratorium Medis Poltekkes Kemenkes Yogyakarta, dilakukan dua metode pewarnaan: konvensional dan Chamber stain. Penilaian dilakukan secara mikroskopis oleh dua Ahli Teknologi Laboratorium Medis. Hasil : Hasil menunjukkan pewarnaan dengan Chamber stain hingga hari ke-4 tetap memberikan kualitas hasil yang baik dan mulai mengalami penurunan pada hari ke-5. Metode ini mampu menghemat penggunaan reagen Giemsa hingga 88,33%, serta menghasilkan pewarnaan yang merata dan bebas pengendapan. Kesimpulan : Pewarnaan Giemsa 15% secara berulang menggunakan Chamber stain efektif dan efisien untuk digunakan. Chamber stain terbukti lebih efisien dalam penggunaan reagen dan waktu dibandingkan metode konvensional, serta mampu menghasilkan kualitas mikroskopis yang setara selama penggunaan berulang dalam batas tertentu.
