Abstract

Background : Xylene is commonly used as a clearing agent in histotechnology due to its effectiveness in tissue clearing, however it is toxic and may pose risk to laboratory personnel and the environment. Olive oil possesses lipophilic properties and contains unsaturated fatty acids, making it a potential safer alternative clearing agent. Heating olive oil may reduce its viscosity and improve tissue penetration. Objective : To determine the effectiveness of olive oil heated at 40˚C as a clearing ageant on the histomorphological quality of Wistar rat liver tissue. Methods: This study used a quasi-experimental design with an intact groups design approach. The research subjects were Wistar rat liver tissues divided into a control group using xylene and a treatment group using olive oil heated at 40˚C. Tissue preparations were processed through histotechnological procedures and stained using Hematoxylin-Eosin (H&E). Histomorphological quality assessment was performed based on nuclear clarity, cytoplasmic clarity, and staining uniformity in five microscopic fields for each slide. Data were analyzed descriptively. Results: Liver tissue preparations cleared with xylene showed a mean quality score of 2.73, while preparations cleared with olive oil heated at 40˚C showed a mean score of 2.70. Histomorphological observations in both groups demonstrated clearly visible nuclei, cytoplasm, sinusoids, and portal triad structures with relatively uniform staining. Mild artifacts were observed in the olive oil group, however, the preparations remained suitable for microscopic evaluation. Conclusion: Olive oil heated at 40˚C has the potential to be used as an alternative clearing agent in the preparation of Wistar rat liver histological slides because it produces good histomorphological quality comparable to xylene. Keywords: olive oil, clearing agent, liver.
