Abstract
ABSTRACT Background: Control material is a material used to monitor the accuracy of a laboratory test. Pooled serum is the waste of human serum after examination. This waste can be utilised as a control material rather than wasted. Pooled serum has similar components with specimens. Based on the similarity of components and ease of obtaining it is stated that pooled serum can be used as an alternative control material. Objective: To determine the homogeneity and stability of pooled serum with the addition of 0.1% sodium azide when used as an alternative control material in total cholesterol examination. Methods: This research is a pre-experimental designs with a research design of one group pretest posttest design. The sample of this study was pooled serum with the addition of 0.1% sodium azide. Data analysis used ISO 13528:2015 homogeneity and stability test formula. Results: Homogeneity test data is declared homogeneous if Ss ≤ 0.3σ. In pooled serum with the addition of 0.1% sodium azide before storage for 8 weeks at 20ºC, the results were 0.936898 ≤ 1.545094403. Then the sample is declared homogeneous and fulfils the criteria. Stability test data is declared stable if |Xr-Yr| ≤ 0.3σ. In pooled serum with the addition of 0.1% sodium azide after being stored for 8 weeks at 20ºC, the result is 1.875 ≥ 1.545. Then the sample is declared unstable because it does not meet the criteria. Conclusion: Total cholesterol levels in pooled serum with the addition of 0.1% sodium azide were homogeneous before storage for 8 weeks at -20ºC and unstable after storage for 8 weeks at 20ºC. Keywords: Homogeneity, stability, pooled serum, cholesterol ABSTRAK Latar Belakang: Bahan kontrol merupakan bahan yang digunakan untuk memantau ketepatan suatu pemeriksaan laboratorium. Pooled serum adalah sisa serum manusia setelah pemeriksaan. Limbah ini dapat dimanfaatkan sebagai bahan kontrol daripada terbuang percuma. Pooled serum ini memiliki kesamaan komponen dengan spesimen. Berdasarkan kesamaan komponen dan kemudahan didapat dinyatakan bahwa pooled serum dapat digunakan sebagai bahan kontrol alternatif. Tujuan: Mengetahui homogenitas dan stabilitas pooled serum dengan penambahan Natrium Azida 0,1% jika digunakan sebagai bahan kontrol alternatif pada pemeriksaan kolesterol total. Metode: Jenis penelitian ini adalah pra eksperimental (pre experimental designs) dengan desain penelitian one group pretest posttest design. Sampel penelitian ini adalah pooled serum dengan penambahan Natrium Azida 0,1%. Analisis data menggunakan rumus uji homogenitas dan stabilitas ISO 13528:2015. Hasil: Data uji homogenitas dinyatakan homogen jika Ss ≤ 0,3σ. Pada pooled serum dengan penambahan Natrium Azida 0,1% sebelum disimpan selama 8 minggu pada suhu 20ºC didapatkan hasil 0,936898 ≤ 1,545094403. Maka sampel dinyatakan homogen dan memenuhi kriteria. Data uji stabilitas dinyatakan stabil jika |Xr-Yr| ≤ 0.3σ. Pada pooled serum dengan penambahan Natrium Azida 0,1% setelah disimpan selama 8 minggu pada suhu 20ºC didapatkan hasil 1,875 ≥ 1,545. Maka sampel dinyatakan tidak stabil karena tidak memenuhi kriteria. Kesimpulan: Kadar kolesterol total pada pooled serum dengan penambahan Natrium Azida 0,1% homogen sebelum disimpan selama 8 minggu pada suhu -20ºC dan tidak stabil setelah disimpan selama 8 minggu pada suhu 20ºC.
