Abstract

Background : Patients with Diabetes Mellitus are often found to have high blood cholesterol levels, this is due to abnormalities of fat metabolism which results in increased free fatty acids in the blood. High cholesterol levels will cause plaque and blockage in blood vessels, resulting in various complications of the disease. Black garlic contains polyphenol antioxidant compounds that increase the activity of super-oxide dismutase and catalase enzymes so that it can reduce oxidative stress, be able to control blood glucose levels, prevent diabetes complications such as hypercholesterolemia and be effective in lowering blood sugar. Purpose : Knowing the effect of black garlic (Black Allium sativum) with a certain composition on the total blood cholesterol levels of diabetic white rats (Rattus novergicus). Method : This research is an experimental laboratory study with a "pre and posttest controlled group design" research design, conducted at the Laboratory of Food and Nutrition Study at PAU, Gadjah Mada University, Yogyakarta. The subjects of this study were 30 male white rats (Rattus norvegicus), Wistar strain, ± 2 months old with a body weight of ± 200 grams. This study used 6 experimental groups, consisting of 1 negative control group, 1 positive control group, and 4 treatment groups. Data analysis used paired t-test, one-way ANOVA test, and post hoc test. Result : The results showed the effect of giving black garlic to decrease total blood cholesterol levels of white rats (Rattus novergicus) who suffer from Diabetes Mellitus (p: 0,000) p <0.01. There is an effect of giving black garlic to decrease total blood cholesterol levels of normal white rats (Rattus novergicus) (p: 0.033) p <0.05. Conclusion : There is an effect of giving black garlic (Black Allium sativum) to a decrease in the total blood cholesterol level of diabetic white rats (Rattus novergicus). Latar Belakang : Penderita Diabetes Mellitus sering didapati kadar kolesterol dalam darahnya tinggi, hal tersebut terjadi karena kelainan metabolisme lemak yang berakibat meningkatnya asam lemak bebas dalam darah. Tingginya kadar kolesterol akan menyebabkan terjadinya plak dan sumbatan di pembuluh darah sehingga mengakibatkan berbagai komplikasi penyakit. Bawang hitam mengandung senyawa antioksidan golongan polifenol yang meningkatkan aktivitas super-oksida dismutase dan enzim katalase sehingga dapat mengurangi stress oksidatif, mampu mengontrol kadar glukosa darah, mencegah komplikasi diabetes seperti hiperkolesterolemia dan efektif dalam menurunkan gula darah sewaktu. Tujuan : Mengetahui pengaruh pemberian bawang hitam (Black Allium sativum) dengan komposisi tertentu terhadap kadar kolesterol darah total tikus putih (Rattus novergicus) yang diabetes. Metode : Penelitian ini adalah penelitian eksperimental laboratorik dengan rancangan penelitian “pre and posttest controlled group design”, dilakukan di Laboratorium Pusat Studi Pangan dan Gizi PAU Universitas Gadjah Mada Yogyakarta. Subjek penelitian ini adalah tikus putih (Rattus norvegicus) jantan sebanyak 30 ekor, strain Wistar, berumur ± 2 bulan dengan berat badan ± 200 gram. Penelitian ini menggunakan 6 kelompok percobaan, terdiri dari 1 kelompok kontrol negative, 1 kelompok kontrol positif, dan 4 kelompok perlakuan. Analisis data menggunakan uji t-berpasangan, uji one-way ANOVA, dan uji post hoc. Hasil : Hasil penelitian menunjukkan adanya pengaruh pemberian bawang hitam terhadap penurunan kadar kolesterol darah total tikus putih (Rattus novergicus) yang menderita Diabetes Mellitus (p : 0,000) p<0,01. Ada pengaruh pemberian bawang hitam terhadap penurunan kadar kolesterol darah total tikus putih (Rattus novergicus) normal (p : 0,033) p<0,05. Kesimpulan : Ada pengaruh pemberian bawang hitam (Black Allium sativum) terhadap penurunan kadar kolesterol darah total tikus putih (Rattus novergicus)Diabetes Mellitus.
